- Abstract
Fluorescent proteins represent an integral component of biological imaging, with a wide variety of techniques and applications. Their photophysics frequently introduce stochastic dark states or “blinking” behavior, a trait that has been functionalized in super resolution imaging. Herein, we present a high throughput, unbiased method for analysis and binarization of single molecule blinking, designed to extract the kinetics of these behaviors. This is done at low irradiances appropriate for biological imaging to directly further fluorescent protein understanding in the context of SOFI and other techniques.
